 我々はゼブラフィッシュにおいて、トランスポゾンTol2を用いた遺伝子トラップ法を確立してきた。今回、我々はトランスポゾンを用いた新しいゲノム改変方法構築のために、ゲノムに組み込まれたTol2の再転移の解析を行った。転移酵素mRNAを微量注入したTol2挿入ホモ2倍体受精卵を成魚に育て、かけあわせ、次世代の魚を得た。40匹の微量注入した魚から新たなGFP発現パターンを持つ10系統が得られた。これらについて分子生物学的解析を行い、ゲノムに組み込まれたTol2が元の場所から高頻度で切り出され、新たな部位に挿入することを明らかにした。再転移により復帰変異体及び新しい遺伝子トラップ系統が作成可能である。

 We have been developing the gene trap method using the Tol2 transposon in zebrafish. To date, we have identified and characterized 102 unique GFP expression patterns. 75% of the genes trapped by the transposon insertions turned out to be previously uncharacterized genes, indicating that the gene trap method is useful to identify novel developmental genes. Currently, chromosomal insertions of Tol2 are created by plasmid injection. To develop a novel method to create insertions, we injected the transposase mRNA into embryos carrying a single transposon insertion in the hoxc cluster and tested whether the integrated genomic copy could be mobilized. 40 injected fish were mated and the F1 progeny was analyzed. In F1 fish, we identified 10 new GFP expression patterns, different from that of the original insertion. Molecular characterization of these fish revealed that the integrated transposon was excised from the original locus efficiently and integrated in new loci located on different chromosomes. Thus, we successfully developed a method for remobilization by which revertants of existing insertions and insertions of new loci can be created efficiently.
